Chemokine Gradient Analysis

Chemokine gradient analysis is the study of how spatial and temporal differences in chemokine concentration guide the movement and positioning of cells. In bioengineering, researchers generate or measure gradients by controlling chemokine transport through diffusion, fluid flow, binding, and degradation, then assess how cells detect concentration changes through chemokine receptors and migrate by chemotaxis. This analysis helps quantify gradient shape, stability, steepness, and cellular response in engineered microenvironments. Applications include designing microfluidic migration assays, modeling immune-cell recruitment, evaluating tissue-engineered scaffolds, and developing systems that regulate inflammation, vascularization, or targeted cell delivery.

The three-dimensional flow chamber device is a novel in vitro technology for the quantitative and step-wise evaluation of the extravasation cascade of cells circulating under conditions of physiological shear stress. The control of chemical and oxygen gradients is essential for cell cultures. This paper reports a polydimethylsiloxane-polycarbonate (PDMS-PC) microfluidic device capable of reliably generating combinations of chemical and oxygen gradients for cell migration studies, which can be practically utilized in biological labs without sophisticated instrumentation.
