Fluorescence Retinal Imaging

Fluorescence retinal imaging is a noninvasive technique that visualizes retinal structures and blood flow by detecting light emitted from fluorescent compounds after excitation. In methods such as fluorescein angiography, an injected dye circulates through retinal vessels, and a fundus camera or scanning laser ophthalmoscope captures its excitation and emission signals over time, revealing perfusion and vascular leakage. In bioengineering, these images support the study of retinal microvascular architecture, blood-retinal barrier function, and disease-related changes. Quantitative image analysis can also guide the development of diagnostic tools, drug-delivery systems, artificial retinas, and other technologies for monitoring retinal health.

The retina shares prominent similarities with the brain and thus represents a unique window to study vasculature and neuronal structure in the brain non-invasively. This protocol describes a method to study dementia using retinal imaging techniques. This method can potentially aid in diagnosis and risk assessment of dementia.

Identification of Mouse Retinal Ganglion Cell Subtypes Through Fluorescent Tracing

Source: Laboissonniere, et. al. Single-cell RNA-Seq of Defined Subsets of Retinal Ganglion Cells. J. Vis. Exp. (2017). This video demonstrates a fluorescent tracing technique to identify retinal ganglion cell (RGC) subtypes in a transgenic mouse retina. The process involves securing the isolated retina in a recording chamber, maintaining cell viability with oxygenated perfusion, and targeting fluorescently labeled RGCs using a pipette with a tracer. By analyzing dendritic morphology and...

Source: Bosco, A., et al. In Vivo Dynamics of Retinal Microglial Activation During Neurodegeneration: Confocal Ophthalmoscopic Imaging and Cell Morphometry in Mouse Glaucoma. J. Vis. Exp. (2015) This video demonstrates in vivo imaging of retinal microglia in a mouse model of glaucoma. A transgenic mouse with fluorescent protein-expressing retinal microglia is positioned under an ophthalmoscope. Infrared illumination is used to locate the optic nerve head (ONH), and the focal plane is...
