Immunocytochemical Studies

Immunocytochemical studies are laboratory methods that use antibodies to identify and visualize specific proteins or other antigens within individual cells, providing spatial information about cellular structure and function. Typically, cells are fixed and permeabilized, then exposed to a primary antibody that binds the target antigen; a labeled secondary antibody produces a fluorescent or enzymatic signal that can be detected by microscopy. In bioengineering, these studies help evaluate cell identity, differentiation, signaling, and responses to biomaterials or engineered microenvironments. By linking molecular markers to cellular location and morphology, immunocytochemistry supports tissue engineering, regenerative medicine, disease modeling, and validation of engineered cell systems.

Regular characterization of induced pluripotent stem cells (iPSCs), to ascertain maintenance of their pluripotent state, is an important step before these cells are used for other applications. The main adherent cell types derived from human muscle are myogenic cells and fibroblasts. Here, cell populations are enriched using magnetic-activated cell sorting based on the CD56 antigen. Subsequent immunolabelling with specific antibodies and use of image analysis techniques allows quantification of cytoplasmic and nuclear characteristics in individual cells.
