Fibroblast Growth Factors

Fibroblast growth factors are a family of signaling proteins that regulate cell behavior, making them important in development, tissue maintenance, and disease biology. They act by binding fibroblast growth factor receptors, activating receptor tyrosine kinase signaling and downstream pathways that influence cell proliferation, survival, migration, and differentiation. In biology, these signals help coordinate embryonic patterning, blood vessel formation, and wound repair, while abnormal fibroblast growth factor activity can contribute to cancer and other disorders. Studying fibroblast growth factors supports research into tissue regeneration, developmental mechanisms, disease progression, and potential therapeutic strategies.

In this study, we present an in vitro cell assay for identifying teratogenic chemicals. The dynamics of fibroblast growth factor (FGF)/serum response factor (SRF) signaling were measured in human induced pluripotent stem (iPS) reporter cells, and the risk of teratogenicity was evaluated by quantifying chemical disruption of this signaling pathway.

Reprogramming Mouse Embryonic Fibroblasts with Transcription Factors to Induce a Hemogenic Program

The protocol described here details the induction of a hemogenic program in mouse embryonic fibroblasts via overexpression of a minimal set of transcription factors. This technology may be translated to the human system to provide platforms for future study of hematopoiesis, hematologic disease, and hematopoietic stem cell transplant.

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.
