Lymphocyte Reconstitution

Lymphocyte reconstitution is the restoration of T-cell, B-cell, and natural killer cell populations after immune depletion, making it an important indicator of immune recovery in biology and medicine. It occurs through new lymphocyte production in the bone marrow and thymus, alongside homeostatic proliferation of surviving cells and maturation driven by cytokine signals and antigen exposure. Researchers monitor lymphocyte counts, subsets, and functional responses to assess recovery after hematopoietic stem cell transplantation, chemotherapy, infection, or other immunosuppressive treatments. These measurements help evaluate susceptibility to infection, vaccination responses, treatment outcomes, and the development of long-term immune competence.

Efforts to isolate the catalytic subunit of telomerase, TERT, in sufficient quantities for structural studies, have been met with limited success for more than a decade. Here, we present methods for the isolation of the recombinant Tribolium castaneum TERT (TcTERT) and the reconstitution of the active T. castaneum telomerase ribonucleoprotein (RNP) complex in vitro.
