Paraquat Rotenone

Paraquat and rotenone are widely studied environmental toxicants: paraquat is a herbicide, whereas rotenone is a plant-derived insecticide. Both can disrupt mitochondrial function, but through different mechanisms. Paraquat undergoes redox cycling, transferring electrons to oxygen and generating reactive oxygen species, while rotenone inhibits electron transfer through mitochondrial complex I, limiting ATP production and promoting oxidative stress. In biology and toxicology, these compounds are used to investigate mitochondrial dysfunction, membrane damage, cell death, and neuronal injury. Their effects also support experimental models of Parkinsonian neurodegeneration, although outcomes depend on exposure level and biological context.

Parkinson's disease has been related to the exposure to pesticides. Here we show a method to deliver pesticides using a gastric tube at the desired concentration and a method to analyze their effect in alpha-synuclein accumulation in the enteric nervous system.

LC-MS Analysis of Human Platelets as a Platform for Studying Mitochondrial Metabolism

Here we show isolated human platelets can be used as an accessible ex vivo model to study metabolic adaptations in response to the complex I inhibitor rotenone. This approach employs isotopic tracing and relative quantification by liquid chromatography-mass spectrometry and can be applied to a variety of study designs.

Levels of the inflammatory cell-signaling molecule nitric oxide (NO) are commonly assayed using Griess reagent. In this protocol, we have created a modified Griess assay utilizing live Drosophila brain tissue in order to detect the secretion of NO in a simple, quantifiable and highly repeatable method.
