Quantitative Angiogenesis Assessment

Quantitative angiogenesis assessment is the measurement of new blood-vessel formation, organization, and growth, providing objective data for studying vascular biology. It typically combines microscopy or imaging with image-analysis methods that quantify features such as vessel length, density, branching, area, and network connectivity under defined experimental conditions. In biology, these measurements help characterize angiogenesis in development, wound repair, cancer, and other disease models, while enabling comparisons between treatments or genetic conditions. By converting visual vessel patterns into reproducible numerical outcomes, quantitative assessment strengthens experimental rigor and supports evaluation of pro-angiogenic and anti-angiogenic interventions.

This protocol demonstrates a fluorescence-based method to visualize the vasculature and to quantify its complexity in Xenopus tropicalis. Blood vessels can be imaged minutes after the injection of a fluorescent dye into the beating heart of an embryo after genetic and/or pharmacological manipulations to study cardiovascular development in vivo.

Assessing the Angiogenesis Effects of a Test Drug in a Rat Model of Ischemic Stroke

2025

Begin with a rat subjected to middle cerebral artery occlusion, a surgical procedure that temporarily blocks blood flow to the brain tissue. This induces an ischemic stroke, leading to brain tissue damage.Next, remove the blockage to restore blood flow.Select rats that exhibit neurological deficits to ensure study relevance. Then, administer the test drug to the experimental group while injecting saline into the other group.Repeat the drug and saline administration. The test drug promotes...

Normal adult vascularized mammalian tissue that lacks physiologic angiogenesis and that has not been exposed to surgical intervention is used to study: (i) the initiation and development of angiogenesis following intraperitoneal administration of test agents; and (ii) modification of angiogenesis following systemic administration of selected test agents.

Preparation Of Neovascular Tissues from Human Glioma Tissues for Quantitative Proteomics Analysis of Tumor Angiogenesis

2026

This protocol successfully prepared sufficient high-quality neovascular tissue from glioma samples using laser-capture microdissection for quantitative proteomics analysis of tumor angiogenesis, overcoming the limitations of studies that traditionally focus mainly on angiogenic factors and providing a large-scale proteomics profile of glioma angiogenesis.

Christopher Hughes: An in vitro model for the Study of Angiogenesis (Interview)

Christopher C.W. Hughes describes the utility of his culture system for studying angiogenesis in vitro. He explains the importance of fibroblasts that secrete a critical, yet unidentified, soluble factor that allow endothelial cells to form vessels in culture that branch, form proper lumens, and undergo anastamosis.
