Semi-intact Preparation

Semi-intact preparation is a biological experimental technique that preserves selected tissue structures and connections while making internal organs or cells accessible for observation and manipulation. Researchers carefully remove surrounding tissues without fully isolating the system, maintaining key anatomical relationships and, when possible, physiological activity during imaging, recording, or stimulation. This approach enables investigation of cell signaling, neural circuits, muscle function, organ physiology, and interactions between tissues under controlled laboratory conditions. By combining the accessibility of an isolated preparation with the organization of an intact organism, semi-intact preparations help link cellular mechanisms to tissue-level function and provide informative models for studying complex biological processes.

Analysis of vestibular hair cell function is complicated by their location deep within the hardest part of the skull, the petrous temporal bone. Most functional hair cell studies have used acutely isolated hair cells. Here we describe a semi-intact preparation of mouse vestibular epithelium for electrophysiological and two-photon microscopy studies.

Optogenetic Perturbation of Neural Activity with Laser Illumination in Semi-intact Drosophila Larvae in Motion

Here we describe a protocol for optogenetic manipulation of motoneuronal activity while monitoring changes in motor output (muscle contraction) in semi-intact Drosophila larvae using lasers within a conventional confocal microscope. This technique enables researchers to achieve local perturbation of neural activity in a few neuromeres to elucidate the dynamics of motor circuits.
