Xenopus Oocytes

Xenopus oocytes are immature frog egg cells widely used as experimental systems in cell and developmental biology because their large size, resilience, and accessible cytoplasm support precise laboratory manipulation. Researchers can microinject RNA, DNA, proteins, or other molecules into individual oocytes, where injected nucleic acids can drive protein expression and allow direct analysis of cellular processes. Oocytes can also be induced to mature and used to study meiosis, signaling, membrane transport, ion channels, and early developmental events. Their reproducibility and experimental flexibility make them valuable for investigating gene function, protein activity, and the mechanisms underlying vertebrate cell biology.

This article describes an enhancement of conventional Voltage-Clamp Fluorometry (VCF) where Fluorescent Unnatural Amino Acids (fUAA) are used instead of maleimide dyes, to probe structural rearrangements in ion channels. The procedure includes Xenopus oocyte DNA injection, RNA/fUAA coinjection, and simultaneous current and fluorescence measurements.

Optimized Analysis of Proteins from Xenopus Oocytes and Embryos by Immunoblotting

2025

This article describes a protocol for analyzing proteins from Xenopus oocytes and embryos by immunoblotting. Collection steps are described, followed by steps corresponding to sample processing, SDS-PAGE, transfer, antibody staining, and imaging. The protocol emphasizes studying translational regulatory protein complexes with endogenous antibodies and antibodies against protein affinity tags.
