Intracellular Electrode

An intracellular electrode is a fine electrical probe used to record voltage changes inside a living cell, providing direct information about membrane potential and cellular excitability. Typically, a sharp microelectrode or patch pipette crosses the cell membrane, while a reference electrode outside the cell measures the voltage difference; the same setup can also inject controlled current and assess how the cell responds. In biology, intracellular recording reveals resting potentials, action potentials, synaptic responses, and ion-channel activity in neurons, muscle cells, and other excitable tissues. These measurements help researchers connect electrical signaling with cellular communication, development, disease mechanisms, and drug effects.

In this protocol a method to measure intracellular protein refolding after heat shock is described. This method can be used to study foldases like molecular chaperones and their co-factors or compounds able to influence their activity. Firefly luciferase activity is used as reporter to measure chaperone refolding activity.

This video demonstrates the recording of electrical signals from single brain neurons using a fine-tipped glass micropipette and a specialized setup. The micropipette is carefully inserted into the rat's brain and is buzzed to penetrate the neuron to record internal neuronal electrical signals.

We demonstrate how changes in the intracellular free calcium concentration and synaptic efficacy can be simultaneously monitored in a ganglion preparation of Aplysia. We image intracellular calcium using a fluorescent dye, Calcium Orange, and induce and monitor synaptic transmission with sharp (intracellular) electrodes.
