Brain Tumor Engraftment

Brain tumor engraftment is the establishment of human or animal brain tumor cells or tissue within a living host, typically to create an in vivo model of tumor growth. During the process, tumor cells or fragments are introduced into a selected brain region or implantation site, where they survive, interact with the surrounding microenvironment, and form a growing mass supported by host blood vessels and signaling pathways. In cancer research, engraftment models help investigators examine tumor progression, invasion, treatment response, and recurrence under biologically relevant conditions, supporting preclinical evaluation of therapies and comparisons between tumor types or patient-derived samples.

Mantle cell lymphoma (MCL) is a difficult to treat B cell disorder and it is equally difficult to establish a xenograft mouse model of primary MCL to study and develop therapeutics. Here, we describe the successful establishment of MCL xenografts in mice to help understand its underlying biology.

The transplantation of cancer cells is an important tool for the identification of cancer mechanisms and therapeutic responses. Current techniques depend on immune-incompetent animals. Here, we describe a method to transplant zebrafish tumor cells into immune-competent embryos for the long-term analysis of tumor cell behavior and in vivo drug responses.

A mouse model of human endoscopic skull base reconstruction has been developed that creates a semipermeable interface between the brain and nose using nasal mucosal grafts. This method allows researchers to study delivery to the central nervous system of high molecular weight therapeutics which are otherwise excluded by the blood-brain barrier when administered systemically.
