Human Fibroblasts

Human fibroblasts are connective-tissue cells that produce and remodel extracellular matrix, helping maintain tissue structure, repair injury, and regulate local cellular environments. They synthesize components such as collagen and fibronectin while responding to biochemical and mechanical signals that alter their proliferation, migration, and secretory activity. In cancer research, fibroblasts are studied as key components of the tumor microenvironment because cancer-associated fibroblasts can reshape the extracellular matrix, release signaling factors, and influence tumor growth, invasion, immune responses, and treatment sensitivity. Cultured fibroblasts and co-culture models help clarify tumor-stroma interactions and identify potential therapeutic targets.

Experimental Generation of Carcinoma-Associated Fibroblasts (CAFs) from Human Mammary Fibroblasts

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Here, a protocol to isolate and establish primary fibroblast/myofibroblast (MF) cultures from frozen gastric, small intestinal, and colonic tissue-yielding cells with a MF phenotype-is presented. These cells express CD90, α-SMA and vimentin. MFs can be used for a variety of functional assays including enzymatic activity and cytokine production.

Although proliferative polyploid cells are necessary to analyze chromosomal instability of polyploid cells, creating such cells from nontransformed human cells is not easy.
