Phosphopeptide Enrichment

Phosphopeptide enrichment is a sample-preparation strategy that selectively concentrates phosphorylated peptides from complex protein digests for mass spectrometric analysis. It commonly uses titanium dioxide or immobilized metal affinity chromatography, in which phosphate groups bind preferentially to metal oxide or positively charged metal ions under controlled chemical conditions before unbound peptides are removed and enriched peptides are eluted. In cancer research, this process improves detection of low-abundance phosphorylation sites that regulate signaling pathways, kinase activity, cell proliferation, and DNA-damage responses. The resulting phosphoproteomic profiles help identify dysregulated networks, characterize tumor biology, and support biomarker or therapeutic-target discovery.

Post-translational modifications (PTMs) change protein structures and functions. Methods for the simultaneous enrichment of multiple PTM types can maximize coverage in analyses. We present a protocol using dual-functional Ti(IV)-immobilized metal affinity chromatography followed by mass spectrometry for the simultaneous enrichment and analysis of protein N-glycosylation and phosphorylation in pancreatic tissues.
