Carbon-11 Radiolabeling

Carbon-11 radiolabeling is a radiochemical technique that replaces a stable carbon atom in a molecule with the positron-emitting isotope carbon-11, enabling molecular imaging and tracing in living systems. Carbon-11 is typically produced in a cyclotron and rapidly converted into reactive labeling agents, such as [11C]carbon dioxide or [11C]methyl iodide, which are incorporated into target compounds through chemical synthesis before radioactive decay. Because carbon-11 has a half-life of about 20 minutes, researchers must optimize reactions for speed, selectivity, and efficient purification. In chemistry and positron emission tomography, the method supports studies of drug distribution, receptor binding, metabolism, and biological processes in real time.

Enhancing Efficiency and Radiolabeling Yields of Carbon-11 Radioligands for Clinical Research Using the Loop Method

This protocol presents a detailed procedure to prepare four carbon-11 radiotracers by way of the “loop method” [11C]methylation. We describe the procedure to synthesize [11C]ER-176, with data for three additional radiotracers: [11C]MRB, [11C]mHED, and [11C]PiB. The loop method provides an efficient synthesis with increased radiochemical yields against traditional reaction vessel [11C]methylation.

This paper describes a method for the measurement of fuel oxidation in Drosophila melanogaster in which trace amounts of specific radiolabeled metabolic substrates are fed to flies. The exhaled radiolabeled CO2 that is a produced from fuel oxidation is collected and measured.
