Density Method

Density methods are analytical approaches used to determine a substance’s density, the ratio of its mass to its volume, an important property for identifying materials and assessing composition in chemistry. They work by measuring mass and volume directly or by observing how a sample displaces a known volume of liquid; for liquids, instruments such as hydrometers relate buoyancy to density. Temperature control is essential because volume and density often change with temperature. These methods support purity testing, concentration measurements, phase characterization, and quality control for solids, liquids, and solutions, while also helping researchers compare substances and monitor chemical processes.

Low-density neutrophils (LDN) increase significantly in several diseases. LDN are usually isolated through cell sorting. We present a practical method to obtain pure and viable LDN. After density-gradient centrifugation of peripheral blood, cells are incubated with magnetic microbeads, and then LDN are separated through magnetic columns.

We describe here a simplified method to culture ultra-low density neurons on glass coverslips in the presence of a high density neuronal feeder layer, which facilitates investigation of specific neuronal-autonomous mechanisms.
