Intracellular Metabolite Detection

Intracellular metabolite detection is the measurement of small molecules within cells to reveal how metabolism changes across physiological and disease states. Methods such as fluorescence-based biosensors, mass spectrometry, and nuclear magnetic resonance identify metabolites through selective molecular recognition, ionization and mass-to-charge analysis, or characteristic magnetic signals, respectively. These approaches can quantify compounds including sugars, amino acids, lipids, and signaling intermediates while preserving information about cellular location and timing in some applications. In chemistry and cell biology, intracellular metabolite detection supports pathway mapping, drug-response analysis, biomarker discovery, and investigation of metabolic regulation.

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.

This manuscript describes a novel technique which allows the detection of intracellular gene expression after endocytosis of fluorescence-labeled nanoparticles directly in live cells. The method does not require manipulation of the cells and is not restricted with respect to the target gene or species.
