Ex Vivo Bone Culture

Ex vivo bone culture is a laboratory method that maintains living bone tissue outside the organism to study its cellular organization, growth, and responses under controlled conditions. Researchers place bone explants in nutrient media that support osteoblasts, osteoclasts, progenitor cells, and the surrounding extracellular matrix, preserving interactions involved in bone formation and remodeling. In developmental biology, this approach helps examine skeletal development, mineralization, tissue patterning, and signaling between bone cells without the complexity of a whole organism. It also provides a controlled platform for testing genetic, molecular, or environmental influences on bone growth and evaluating candidate treatments before in vivo studies.

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.
