Non Colony Monolayer

A non-colony monolayer is a cell culture in which adherent cells grow as a dispersed, continuous layer across a laboratory surface rather than forming distinct colonies. Cells attach to a treated substrate, spread, and proliferate within a controlled medium, allowing researchers to observe cell behavior across an accessible two-dimensional field. In developmental biology, this format supports the study of cell adhesion, migration, proliferation, differentiation, and responses to signaling cues. Its uniform layout also facilitates microscopy, experimental manipulation, and comparison of developmental phenotypes, providing a practical model for examining how cells organize and change during tissue formation.

We present a procedure for forming a poly(ethylene glycol) self-assembled monolayer (PEG-SAM) on a silicon substrate with gold microelectrodes. The PEG-SAM is formed in a single step and prevents biofouling on silicon and gold surfaces. Electrophoresis is then used for patterning biomolecules down to the nanoscale.

A method for embedding yeast colonies allowing sectioning for light and electron microscopy. This protocol allows determination of the distribution of sporulated cells and pseudohyphal cells within colonies providing a new tool toward understanding the organization of cell types within a fungal community.
