Pre-implantation Development

Pre-implantation development is the early phase of embryogenesis in which a fertilized egg undergoes coordinated divisions and cellular specialization before attaching to the uterine lining. After fertilization, cleavage divisions increase cell number without substantially increasing embryo size; compaction then organizes cells into a morula, followed by formation of a fluid-filled blastocyst with distinct inner cell mass and trophectoderm lineages. Studying this process helps explain how embryos establish their first body plan, activate developmental gene networks, and prepare for implantation. It also supports research in developmental biology, in vitro fertilization, embryo culture, infertility, and the causes of early developmental failure.

Microarray technology allows quantitative measurement and gene expression profiling of transcripts on a genome-wide basis. Therefore, this protocol provides an optimized technical procedure in a two-color custom made bovine array using Day 7 bovine embryos to demonstrate the feasibility of using low amount of total RNA.

In eukaryotic cells, transcripts made by RNA polymerase are modified and processed before exiting the nucleus. Unprocessed RNA is called precursor mRNA or pre-mRNA to distinguish it from mature mRNA.
