Avian Transgenesis

Avian transgenesis is the deliberate introduction and inheritance of foreign or modified genetic material in birds, providing a way to connect gene activity with embryonic development and organismal traits. In a typical strategy, primordial germ cells or early embryos receive a transgene through delivery systems such as viral vectors or genome-editing tools; modified germ cells can then contribute to the developing embryo and transmit the altered sequence to offspring. In developmental biology, these models help test gene function, regulatory elements, cell-lineage decisions, and tissue formation in vivo. They also support studies of avian disease, reproduction, evolution, and the production of birds with precisely altered characteristics.

Transgenic manipulations and genome editing are critical for functionally testing the roles of genes and cis-regulatory elements. Here a detailed microinjection protocol for the generation of genomic modifications (including Tol2-mediated fluorescent reporter transgene constructs, TALENs, and CRISPRs) is presented for the emergent model fish, the threespine stickleback.

RNAi Mediated Gene Knockdown and Transgenesis by Microinjection in the Necromenic Nematode Pristionchus pacificus

In model organisms, transgenesis can manipulate gene functions while RNAi can knockdown specific mRNA transcripts 1-2. This protocol aims to illustrate the techniques needed to introduce stably transmitted DNA and transient double stranded RNA into the necromenic nematode Pristionchus pacificus for studies in evolutionary, developmental, and behavioral biology.

Determining the Minimum Inhibitory Concentration of Enrofloxacin Against Avian Pathogenic E. coli

Begin with a suspension of avian pathogenic E. coli or APEC in growth media.APEC is a Gram-negative bacterium that infects poultry by entering through the respiratory tract. After breaching the mucosal layer, APEC adhere to epithelial cells, multiply locally, and disrupt the epithelial barrier to enter the bloodstream. The bacteria then spread to multiple organs, causing systemic infection.Add an equal volume of the bacterial suspension to each well of a multi-well plate containing increasing...
