Stem Cell Regeneration

Stem cell regeneration is the biological process by which stem cells maintain their population and generate specialized cells to restore or build tissues. It depends on self-renewal, controlled cell division, and differentiation, guided by signals from the surrounding cellular niche, developmental pathways, and tissue injury. In developmental biology, these mechanisms help explain how tissues form, grow, and maintain their organization throughout life. Studying stem cell regeneration supports disease modeling, tissue repair research, and regenerative medicine, while also revealing how disrupted signaling can contribute to developmental disorders and poor healing.

Tissue Regeneration with Somatic Stem Cells

Somatic or adult stem cells, like embryonic stem cells, are capable of self-renewal but demonstrate a restricted differentiation potential. Nonetheless, these cells are crucial to homeostatic processes and play an important role in tissue repair. By studying and manipulating this cell population, scientist may be able to develop new regenerative therapies for injuries and diseases. This video first defines somatic stem cells, and then explores the role these cells play in tissue regeneration.

Use of Human Perivascular Stem Cells for Bone Regeneration

Human perivascular stem cells (PSCs) are a novel stem cell class for skeletal tissue regeneration similar to mesenchymal stem cells (MSCs). PSCs can be isolated by FACS (fluorescence activated cell sorting) from adipose tissue procured during standard liposuction procedures, then combined with an osteoinductive scaffold to achieve bone formation in vivo.

Induced pluripotent stem cell (iPSC)-derived myogenic progenitors are promising candidates for cell therapy strategies to treat muscular dystrophies. This protocol describes transplantation and functional measurements required to evaluate the engraftment and differentiation of iPSC-derived mesoangioblasts (a type of muscle progenitors) in mouse models of acute and chronic muscle regeneration.

Chondrogenesis from stem cells requires fine tuning the culture conditions. Here, we present a magnetic approach for condensing cells, an essential step to initiate chondrogenesis. In addition, we show that dynamic maturation in a bioreactor applies mechanical stimulation to the cellular constructs and enhances cartilaginous extracellular matrix production.
