Microbiome Composition Analysis

Microbiome composition analysis is the study of which microorganisms are present in a microbial community and their relative abundances, providing a way to characterize environmental ecosystems. It typically involves collecting soil, water, or sediment, extracting community DNA, sequencing taxonomic marker genes such as 16S rRNA, or sequencing total community DNA, and assigning sequence reads to taxa with bioinformatics. Researchers use these profiles to compare diversity and community structure across habitats or changing conditions, helping assess ecosystem function, environmental disturbance, and microbial contributions to nutrient cycling.

A method for the determination of fatty acid content and composition in microalgae based on mechanical cell disruption, solvent based lipid extraction, transesterification, and quantification and identification of fatty acids using gas chromatography is described. A tripentadecanoin internal standard is used to compensate for the possible losses during extraction and incomplete transesterification.

Lipid polyesters constitute the structural components of two cell wall modifications, the plant cuticle and suberin-containing diffusion barriers. In this video, we describe a method to depolymerize cutin from whole delipidated leaves. The method can be applied to investigating mutants compromised in either cutin or suberin biosynthesis.

Plant biomass is a major carbon-neutral renewable resource that could be used for the production of biofuels. Plant biomass consists mainly of cell walls, a structurally complex composite material termed lignocellulosics. Here we describe a protocol for a comprehensive analysis of the content and composition of the polyphenolic lignin.
