Macropinocytosis Growth

Macropinocytosis growth describes how cells use macropinocytosis, an actin-dependent form of endocytosis, to internalize extracellular fluid and dissolved nutrients in large vesicles called macropinosomes. During this process, plasma-membrane ruffling encloses surrounding fluid, and subsequent macropinosome maturation can release amino acids and other building blocks for cellular growth. In genetics, studying macropinocytosis growth helps reveal how genes controlling actin remodeling, membrane trafficking, and nutrient metabolism influence proliferation and survival. Measuring fluid uptake or disrupting candidate genes can clarify pathway function in development and disease, including tumors that exploit macropinocytosis to support growth under nutrient stress.

Macropinocytosis, large-scale non-specific fluid uptake, is important in many areas of clinical biology including immunology, infection, cancer, and neurodegenerative diseases. Here, existing techniques have been adapted to allow high-throughput, single-cell resolution measurement of macropinocytosis in the macropinocytosis model organism Dictyostelium discoideum using flow cytometry.
