Shrna Transduction

shRNA transduction is a gene-delivery method that introduces DNA encoding a short hairpin RNA (shRNA) into cells to reduce expression of a selected gene. Typically, a viral vector carries the shRNA expression cassette into target cells, where the hairpin is transcribed, processed into small interfering RNA, and loaded into the RNA-induced silencing complex (RISC); the guide strand directs sequence-specific degradation or suppression of complementary messenger RNA. In genetics research, this approach enables stable or transient gene knockdown, allowing investigators to test gene function, model disease-related pathways, and evaluate phenotypic effects in cultured cells or experimental organisms. Careful vector selection and controls help distinguish specific silencing from delivery-related effects.

Transduction

Among the three main modes of HGT—transformation, conjugation, and transduction—transduction is unique in that it is mediated by bacteriophages, or bacterial viruses.Transduction occurs in two ways. Generalized transduction occurs during the lytic cycle of a bacteriophage infection. In this process, bacteriophages infect bacterial cells, replicate within them, and ultimately cause cell lysis, releasing newly assembled virions. Occasionally, random fragments of the bacterial genome are...

(2019).This video demonstrates viral transduction via in-utero injection in mouse embryos. A pregnant mouse is prepared for the surgery, and a midline incision is made to expose the uterine horns. A mixture of a virus containing the desired gene and a tracking dye is injected into the embryos within the uterine sacs at the...
