Three Prime Utr Deletion

Three-prime untranslated region (3′ UTR) deletion is a genetic alteration that removes noncoding sequence downstream of a gene’s protein-coding region, helping researchers determine how this region regulates gene expression. The deletion can eliminate binding sites for microRNAs and RNA-binding proteins, as well as polyadenylation or other regulatory signals, thereby changing messenger RNA stability, localization, processing, or translation without directly altering the encoded protein. In genetics, 3′ UTR deletion experiments support functional analysis of cis-regulatory elements, interpretation of regulatory variants, and investigation of how post-transcriptional control contributes to development, disease, and gene-expression phenotypes.

Gene deletion mutants generated through homologous recombination are the gold standard for gene function studies. The OSCAR (One Step Construction of Agrobacterium-Recombination-ready-plasmids) method for rapid generation of deletion constructs is described. Agrobacterium mediated fungal transformation follows. Finally, a PCR based confirmation method of gene deletions in fungal transformants is presented.
