Cd11b Cd45 Markers

CD11b and CD45 markers are cell-surface proteins used to identify and characterize leukocytes, especially myeloid immune cells, in immunology and infection research. CD45, a protein tyrosine phosphatase found on nearly all nucleated hematopoietic cells, establishes leukocyte identity, while CD11b, the integrin αM subunit, contributes to adhesion, migration, and interactions with complement-coated targets. Fluorescent antibodies against these markers enable flow cytometry or microscopy to distinguish monocytes, macrophages, neutrophils, and other immune populations based on marker expression and intensity. Their combined use helps quantify immune-cell recruitment, activation, and tissue infiltration during infection, inflammation, and immune-response studies.

Here, we present a protocol to isolate microglia from postnatal mouse pups (day 1) for in vitro experimentation. This improvised method of isolation generates both high yield and purity, a significant advantage over alternate methods that allows broad range experimentation for the purposes of elucidating microglial biology.

Liver injuries are accompanied by progenitor cell expansion that represents a heterogeneous cell population. Novel classification of this cellular compartment allows for the distinguishing of multiple subsets. The method described here illustrates the flow cytometry analysis and high purity isolation of various subsets that can be used for further assays.
