Conjunctiva Isolation

Conjunctiva isolation is a laboratory technique used to obtain conjunctival tissue or cells from the ocular surface for controlled biological analysis. The procedure typically involves carefully separating the conjunctiva from adjacent eye tissues, preserving the sample under sterile conditions, and, when needed, processing it into tissue sections or a cell suspension for downstream assays. In immunology and infection research, isolated conjunctival material helps investigators examine epithelial barrier function, resident and recruited immune cells, inflammatory responses, and interactions with infectious agents. These preparations support microscopy, molecular analysis, and cell-based experiments that clarify how the ocular surface detects and responds to disease.

The exposed normal ocular surface consists of cornea and conjunctiva. Epithelial cells, goblet cells and immune cells are present in the conjunctiva. Here, a non-invasive, technique of impression cytology is described using an impression cytology device and flow cytometry to analyze immune cells in the conjunctiva.

Presented here is a multiphoton microscopic platform for live mouse ocular surface imaging. Fluorescent transgenic mouse enables the visualization of cell nuclei, cell membranes, nerve fibers and capillaries within the ocular surface. Non-linear second harmonic generation signals derived from collagenous structures provide label-free imaging for stromal architectures.

We describe the isolation of cardiac extracellular matrix from C57Bl/6J control mice, tight-skin mice, and tight-skin mice treated with the IRF5 inhibitory peptide. We also describe the vasodilation studies on the isolated vessels from C57Bl/6J, tight-skin mice and tight-skin mice treated with the IRF5 inhibitory peptide.
