Hamster Model

A hamster model is a living research system that uses hamsters to investigate biological processes, disease development, and responses to treatment. In immunology and infection research, scientists introduce a pathogen or experimental antigen and monitor how pathogen replication, tissue damage, innate defenses, antibody production, and other adaptive immune responses change over time. These models can reveal links between immune activity and clinical signs while enabling analysis of transmission, disease severity, and protective immunity. Hamster models therefore support evaluation of vaccines, therapeutics, and infection-control strategies, helping researchers connect cellular and molecular findings with outcomes in an intact organism.

Optimization of the experimental hamster model for cutaneous leishmaniasis by intradermal injection of Leishmania promastigotes at the dorsal skin. This approach is useful during inoculation, follow-up, characterization of lesions, application of treatments and obtaining of clinical samples. Locomotion, search for food and water, play and social activities are preserved.

Pronuclear (PN) injection of the clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated protein-9 nuclease (CRISPR/Cas9) system is a highly efficient method for producing genetically engineered golden Syrian hamsters. Herein, we describe the detailed PN injection protocol for the production of gene knockout hamsters with the CRISPR/Cas9 system.

In this video-article we present a step-by-step demonstration on how to collect and cryopreserve hamster oocytes with high post-thaw survival rates. The same procedure can also be applied to successfully freeze and thaw mouse embryos at different stages of preimplantation development.

A Technique to Purify Monoclonal Antibodies Produced by Chinese Hamster Ovary Cells

2025

This video demonstrates an assay for purifying monoclonal antibodies from cell culture fluid harvested from Chinese Hamster Ovarian cells. It employs a porous glass resin with immobilized protein A, which binds to the Fc region of the antibodies in the sample, effectively separating them from contaminants. The use of a low-pH elution buffer weakens the interaction, facilitating the retrieval of purified antibodies.
