Peptide Library Screening

Peptide library screening is a systematic method for testing large collections of short amino acid sequences to identify peptides with a desired biological activity or molecular interaction. Libraries may contain overlapping, randomized, or modified peptides, which are presented to antibodies, receptors, enzymes, or cells and evaluated through binding or functional assays; signal strength links sequence to activity. In immunology and infection research, this approach helps map antigenic epitopes recognized by antibodies or T cells, characterize pathogen-host interactions, and identify sequences that modulate immune responses. These findings support diagnostic assay development, vaccine design, immune monitoring, and the discovery of peptide-based therapeutics, while sequence-level data can guide further validation in cellular or animal models.

Peptide tertiary amides (PTAs) are a superfamily of peptidomimetics that include but are not limited to peptides, peptoids and N-methylated peptides. Here we describe a synthetic method which combines both split-and-pool and sub-monomer strategies to synthesize a one-bead one-compound library of PTAs.

Bacterial Inner-membrane Display for Screening a Library of Antibody Fragments

We provide a method to simultaneously screen a library of antibody fragments for binding affinity and cytoplasmic solubility by using the Escherichia coli twin-arginine translocation pathway, which has an inherent quality control mechanism for intracellular protein folding, to display the antibody fragments on the inner membrane.

Herein, we present a detailed protocol for screening small peptides that bind to FGFR2 using a phage display peptide library. We further analyze the affinity of the selected peptides toward FGFR2 in vitro and its ability to suppress cell proliferation.

This protocol describes a high throughput screen for cellulolytic activity from a metagenomic library expressed in Escherichia coli. The screen is solution based and highly automated, and uses one-pot chemistry in 384 well microplates with the final readout as an absorbance measurement.
