T Cell Subset Differentiation

T cell subset differentiation is the process by which naïve T lymphocytes develop into specialized populations with distinct immune functions, shaping responses to infection and disease. Following antigen recognition through the T cell receptor, costimulatory signals and the surrounding cytokine environment activate lineage-defining transcriptional programs that guide cells toward helper, regulatory, cytotoxic, or memory states. These subsets coordinate pathogen clearance, limit excessive inflammation, and preserve long-term immune protection. Understanding their differentiation supports research into host defense, vaccination, autoimmune disease, immunodeficiency, and cancer immunotherapy, where redirecting T cell fate or function may improve treatment outcomes.

Naïve CD4+ T cells polarize to various subsets depending on the environment at the time of activation. The differentiation of naïve CD4+ T cells to various effector subsets can be achieved in vitro through the addition of T cell receptor stimuli and specific cytokine signals.

Isolation of Precursor B-cell Subsets from Umbilical Cord Blood

Here we describe a protocol for isolating subsets of precursor B-cells from umbilical cord blood. A sufficient quantity and quality of nucleic acids may be extracted from the cells and used in subsequent assays utilizing DNA or RNA.

Distinct dendritic cell subsets exist as rare populations in lymphoid organs, and therefore are challenging to isolate in sufficient numbers and purity for immunological experiments. Here we describe a high efficiency, high yield method for isolation of all of the currently known major subsets of mouse splenic dendritic cells.

This video demonstrates a flow cytometry-based technique using two fluorochromes to identify CD4+ and CD8+ T cells, γδ T cells, B cells, natural killer (NK) cells, and monocytes in human peripheral blood mononuclear cells (PBMCs). Upon incubating the cells with fluorochrome-conjugated antibodies against the surface markers, the cell subsets are identified using an optimal flow cytometric gating strategy.
