Cardiac Function Assessment

Cardiac function assessment is the systematic evaluation of how effectively the heart generates electrical activity, contracts, pumps blood, and maintains adequate circulation. It uses complementary measurements such as electrocardiography to record electrical conduction, echocardiography to visualize chamber motion and blood flow, and blood pressure or cardiac biomarkers to assess hemodynamic performance and myocardial injury. Clinicians apply these findings to identify arrhythmias, valve disease, heart failure, and ischemia while tracking response to treatment. In medicine, integrating test results with symptoms and physical examination supports timely diagnosis, risk stratification, and individualized care, while providing objective endpoints for research and clinical trials.

Langendorff-mode isolated heart perfusion, in conjunction with 31P NMR spectroscopy, combines the fields of biochemistry and physiology into one experiment. The protocol allows for the dynamic measurement of high energy phosphate content and turnover in the heart while concurrently monitoring physiologic function. When performed correctly, this is a valuable technique in the assessment of cardiac energetics.

This protocol aims to describe step-by-step the technique of extraction and assessment of cardiac function using skinned cardiomyocytes. This methodology allows measurement and acutemodulation of myofilament function using small frozen biopsies that can be collected from different cardiac locations, from mice to men.

Isolation of Functional Cardiac Immune Cells

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.
