Diabetic Rabbit Model

A diabetic rabbit model is a laboratory rabbit with experimentally induced or naturally occurring diabetes, used to study disease mechanisms, treatments, and complications in a whole-animal system. Diabetes is commonly produced through chemical destruction of pancreatic beta cells, which reduces insulin secretion and causes sustained hyperglycemia; researchers then monitor blood glucose, insulin responses, vascular changes, and tissue injury. Because rabbits provide sufficient blood and tissue samples while developing measurable metabolic and vascular abnormalities, this model supports studies of glucose regulation, diabetic wound healing, cardiovascular disease, and drug efficacy. Findings can help evaluate therapeutic strategies before clinical testing, while highlighting differences between animal and human physiology.

We describe a surgical procedure used to induce peripheral ischemia in rabbits with hyperlipidemia and diabetes. This surgery acts as a preclinical model for conditions experienced in peripheral artery disease in patients. Angiography is also described as a means to measure the extent of introduced ischemia and recovery of perfusion.

Anesthesia and Monitoring in a Rabbit Model

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Anesthesia is an essential component of rabbit surgical and experimental procedures because it prevents pain, reduces stress responses, and helps immobilize the animal during handling and manipulation when general anesthesia is...

Metabolic memory is the phenomenon by which diabetic complications persist and progress unimpeded even after euglycemia is achieved pharmaceutically. Here we describe a diabetes mellitus zebrafish model which is unique in that it allows for the examination of the mitotically transmissible epigenetic components of metabolic memory in vivo.

Our protocol was developed to cleanly and easily deliver islets or cells under the kidney capsule of mice. Cells are concentrated into pellets in the final tubing used for transplanting the cells under the kidney capsule. The ease of this technique reduces stress to the cells and the mouse.
