DMM-induced osteoarthritis is a preclinical mouse model of post-traumatic knee osteoarthritis produced by destabilization of the medial meniscus, enabling controlled study of disease development. In this surgical technique, the medial meniscotibial ligament is transected, increasing medial meniscus mobility and altering load distribution across the knee; progressive joint instability leads to cartilage degeneration, subchondral bone changes, and synovial inflammation. Researchers use the model to investigate osteoarthritis mechanisms, evaluate imaging and molecular biomarkers, and test potential disease-modifying treatments. Its reproducibility and defined injury mechanism make it valuable for connecting joint biomechanics with structural and inflammatory outcomes in medicine.

Establishment of a Surgically-induced Model in Mice to Investigate the Protective Role of Progranulin in Osteoarthritis

We describe a protocol for the destabilization of the medial meniscus (DMM) model in mice, an effective tool for osteoarthritis (OA) research. In addition, we have demonstrated that deficiency of progranulincan exaggerate OA development and progression by using this model, indicating that progranulin plays a protective role in the pathogenesis of OA.
