Dual-mode Imaging System

A dual-mode imaging system is a medical imaging platform that combines two complementary imaging modalities to provide information that neither modality can supply alone. It works by acquiring data through distinct physical principles, such as anatomical structure and molecular or functional contrast, then aligning or integrating the resulting images through spatial registration and coordinated visualization. This combined view can support lesion localization, diagnosis, treatment planning, and image-guided intervention while helping clinicians relate biological activity to tissue anatomy. Dual-mode systems are therefore valuable in translational research, where multimodal measurements can improve characterization of disease and guide more precise, personalized care.

Dual camera emission splitting systems for two-color fluorescence microscopy generate real-time image sequences with exceptional optical and temporal resolution, a requirement of certain live cell assays including parallel plate flow chamber adhesion assays. When software is employed to merge images from simultaneously acquired emission channels, pseudocolored image sequences are produced.

Here we present a practical guide of building an integrated microscopy system, which merges conventional epi-fluorescent imaging, single-molecule detection-based super-resolution imaging, and multi-color single-molecule detection, including single-molecule fluorescence resonance energy transfer imaging, into one set-up in a cost-efficient way.
