Epithelial Enrichment

Epithelial enrichment is the process of increasing the proportion of epithelial cells in a mixed biological sample, enabling more focused analysis of tissues that line body surfaces and organs. It typically relies on differences between epithelial and non-epithelial cells, such as cell-surface markers, adhesion properties, or physical characteristics, to selectively retain or isolate the desired population. In medicine, enriched epithelial preparations support histopathology, molecular profiling, infection studies, and research on cancer and tissue injury. Improving sample purity can strengthen measurements of epithelial-specific gene expression and cellular behavior, helping researchers characterize disease mechanisms and evaluate potential diagnostic or therapeutic strategies.

This article provides a detailed methodology for tissue dissociation and cellular fractionation approaches allowing enrichment of viable epithelial cells from proximal and distal regions of the human lung. Herein these approaches are applied for the functional analysis of lung epithelial progenitor cells through the use of 3D organoids culture models.
