Injection Site Assessment

Injection site assessment is the systematic examination of skin and underlying tissue before or after an injection to identify local reactions and support safe medication delivery. Clinicians inspect and palpate the site for redness, swelling, warmth, pain, induration, bruising, drainage, or changes in tissue integrity, while considering the injection route and medication administered. Documenting findings and comparing them over time helps distinguish expected, self-limited irritation from complications such as infection, hematoma, or tissue injury. This assessment informs site selection, patient education, follow-up, and timely intervention, improving medication safety across vaccination, therapeutic injection, and clinical care settings.

The method described here utilizes direct injection of entomopathogenic bacteria into the hemocoel of Manduca sexta insect larvae. M. sexta is a commercially available and well-studied insect. Thus, this method represents a simple approach to analyzing host-bacterial interactions from the perspective of one or both partners.

MicroRNAs have significant roles in brain structure and function. Here we describe a method to enforce hippocampal miRNA over-expression using stereotactic injection of an engineered miRNA-expressing lentivirus. This approach can serve as a relatively rapid way to assess the in vivo effects of over-expressed miRNAs in specific brain regions.

A method by which gene expression in the neural tube can be downregulated in a cell type-specific, traceable manner is described. We demonstrate how in ovo electroporation of microRNA-based plasmids that elicit spatiotemporally controlled RNA interference can be used to investigate commissural axon guidance in the developing neural tube.
