Muscle Inflammation

Muscle inflammation is an immune response in skeletal muscle that helps address tissue injury, infection, or other cellular stress, but persistent inflammation can impair muscle function and contribute to disease. Following damage, inflammatory signals recruit immune cells, increase local blood flow, and stimulate the release of mediators that remove damaged components and initiate tissue repair. In medicine, assessing muscle inflammation supports the evaluation of conditions such as myositis, autoimmune disease, and exercise-related injury through clinical findings, laboratory tests, imaging, and sometimes biopsy. Understanding these mechanisms informs diagnosis, treatment selection, rehabilitation, and research into therapies that restore muscle health while limiting chronic tissue damage.

This protocol describes methods for inducing calcification of cultured primary vascular smooth muscle cells and for quantifying calcification and macrophage burden in animal aortas using near-infrared fluorescence imaging.

We describe a non-invasive imaging method for distinguishing inflammatory stages. Systemic delivery of luminol reveals areas of acute inflammation dependent upon MPO activity in neutrophils. In contrast, injection of lucigenin allows for visualization of chronic inflammation dependent upon Phox activity in macrophages.

Subcutaneous Injection of Bacterial Antigen to Induce Systemic Inflammation in a Murine Model

This video demonstrates the induction of systemic inflammation in an anesthetized mouse. The injection of mycobacterial antigen-adjuvant depots triggers cytokine release and localized inflammation. Lymphatic vessels open, enabling immune cell entry into lymph nodes and activation of T cells, which release pro-inflammatory cytokines, leading to widespread inflammation.
