Nerve Lesion Models

Nerve lesion models are experimental systems that reproduce defined injuries to peripheral or central nerves, enabling researchers to study neural damage, repair, and functional loss. These models commonly use procedures such as nerve crush, transection, or compression to disrupt axons, myelin, or supporting tissues, triggering processes including Wallerian degeneration, inflammation, and axonal regeneration. In medicine, nerve lesion models help clarify injury mechanisms, evaluate nerve grafts, biomaterials, rehabilitation strategies, and drug treatments, and measure outcomes such as conduction, sensory recovery, and motor function. Carefully selected lesion models therefore support translational research aimed at improving diagnosis, treatment, and recovery after nerve injury.

A Human Ex Vivo Atherosclerotic Plaque Model to Study Lesion Biology

Atherosclerosis is a chronic inflammatory process. This manuscript illustrates an easy to use ex vivo model to investigate fresh carotid or coronary artery plaques. The ex vivo model allows for the investigation of potential substances on the inflammatory milieu in human atherosclerotic lesions and results can be analyzed by various methods.

Focal demyelination is induced in the optic nerve using lysolecithin microinjection. Visual evoked potentials are recorded via skull electrodes implanted over the visual cortex to examine the signal conduction along the visual pathway in vivo. Parkinson disease is caused by loss of dopaminergic innervation to the striatum, which can be experimentally induced by 6-OH-dopamine. We describe how to perform a stereotaxic lesion and to monitor apomorphine-induced rotational behavior in mice. This model is useful and reliable for testing new therapies for Parkinson disease.
