Quantitative Morphology

Quantitative morphology is the measurement and statistical analysis of biological shape, size, and structural organization, transforming visual features into reproducible numerical data. In medicine, it typically uses microscopy or medical imaging, image segmentation, and computational descriptors to quantify cell, tissue, or organ characteristics under defined conditions. These measurements can reveal subtle morphological changes associated with disease, treatment response, or disease progression and support objective comparisons among patients, specimens, or experimental groups. By complementing qualitative examination, quantitative morphology strengthens diagnostic research, biomarker development, and the evaluation of pathological processes.

This study outlines quantitative measurements of synaptic size and localization, muscle morphology, and mitochondrial shape in C. elegans using freely available image processing tools. This approach allows future studies in C. elegans to quantitatively compare the extent of tissue and organelle structural changes as a result of genetic mutations.

Mapping Microglial Parameters Software (MMPS) is a user-friendly analysis pipeline that requires no coding expertise and semi-automates single-cell microglial morphology analysis from immunofluorescence images. Validated in a rodent lipopolysaccharide-induced neuroinflammation model, MMPS reproducibly detects activation-associated morphological changes and enables standardized, scalable microglial phenotyping.

Whole-mount diaphragm immunohistochemistry assesses morphological innervation at individual neuromuscular junctions. The goal of this protocol is to demonstrate how these two powerful methodologies can be used in various rodent models of spinal cord disease.
