Rat Lung Ischemia

Rat lung ischemia is a condition or experimental model in which blood flow and oxygen delivery to lung tissue are reduced or interrupted, making it useful for studying pulmonary injury. Ischemia limits oxygen-dependent cellular metabolism, while subsequent restoration of circulation can trigger ischemia-reperfusion injury through oxidative stress, inflammation, and disruption of the alveolar-capillary barrier. In medicine, rat lung ischemia models help researchers examine changes in pulmonary function, vascular responses, and tissue damage under controlled conditions. Findings can inform research on lung transplantation, pulmonary vascular disorders, surgical interventions, and strategies to protect lung tissue during periods of inadequate perfusion.

A Model of Acute Lung Injury Following Visceral Ischemia-Reperfusion by Supra-Coeliac Aortic Cross Clamping in Rats

This study describes a model of acute lung injury following visceral ischemia-reperfusion during supra-celiac aortic cross-clamping via laparotomy in rats. It enables the investigation of both the clinical and biological effects of aortic clamping as well as the impact of ischemia-reperfusion on underlying organs and remote lung injuries.

The lung is perfused by both the systemic bronchial artery and pulmonary arteries. In most lung pathologies, it is the smaller systemic vasculature that shows robust neovascularization. Cessation of pulmonary blood flow promotes brisk bronchial angiogenesis. We provide surgical details of inducing left pulmonary artery ischemia that promotes bronchial neovascularization.

Here, we describe the process of harvesting the beating heart from a rat via in situ aortic cannulation, Langendorff perfusion of the heart, simulated ischemia-reperfusion injury, and infarct staining to confirm the extent of ischemic insult.

Ex-Vivo Lung Perfusion (EVLP) has allowed lung transplantation in humans to become more readily available by enabling the ability to assess organs and expand the donor pool. Here, we describe the development of a rat EVLP program and refinements that allow for a reproducible model for future expansion.
