Rfp-labeled Tumor Cells

RFP-labeled tumor cells are cancer cells engineered to express red fluorescent protein (RFP), providing a visible marker for tracking tumor behavior in research and medicine. When illuminated with the appropriate excitation wavelength, RFP absorbs light and emits red fluorescence, allowing labeled cells to be detected by fluorescence microscopy or imaging systems without relying solely on tissue appearance. These cells support studies of tumor growth, migration, invasion, metastasis, and treatment response in culture and experimental models. By enabling researchers to follow cancer cells over time, RFP labeling improves visualization of disease progression and helps evaluate therapeutic strategies.

We describe a method for stable labeling of patient-derived xenografts (PDXs) with lentiviral particles expressing green-fluorescent protein and luciferase reporters. This method allows for tracking the growth of PDXs at the primary site, as well as detecting spontaneous and experimental metastases using in vivo imaging systems.

2011

Limiting dilution cell transplantation assays are used to determine the frequency of tumor-propagating cells. This protocol describes a method for generating syngeneic zebrafish that develop fluorescently-labeled leukemia and details how to isolate and transplant these leukemia cells at limiting dilution into the peritoneal cavity of adult zebrafish.
