Testis Capsule Injection

Testis Capsule Injection is a microsurgical technique for delivering cells, genes, drugs, or other experimental materials into the testis beneath its outer tunica albuginea. By guiding a fine needle through the capsule and controlling injection pressure and volume, researchers can distribute material within testicular tissue while limiting leakage and mechanical injury. In medicine, this approach supports studies of spermatogenesis, testicular development, reproductive disease, and localized therapeutic or gene delivery. It provides a targeted experimental route for evaluating how candidate interventions affect testicular function and may inform future treatments for male infertility and other testicular disorders.

Teratoma Generation in the Testis Capsule

Human pluripotent stem cells (hPSCs) have the potential to treat a myriad of different diseases. The utility of these cells lies in the fact that they can differentiate into any cell type in the body. Here we describe the teratoma assay, which is used to demonstrate the pluripotence of hPSCs.

Germ Cell Transplantation and Testis Tissue Xenografting in Mice

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Testis tissue xenografting reproduces testis development and spermatogenesis of various donor species in recipient mice.

By centrifuging a transposon mutant library through the gradient, bacteria with different capsule densities localize to distinct layers. The separated fractions can then be collected for downstream analysis.

Transplantation of Pancreatic Islets Into the Kidney Capsule of Diabetic Mice

Our protocol was developed to cleanly and easily deliver islets or cells under the kidney capsule of mice. Cells are concentrated into pellets in the final tubing used for transplanting the cells under the kidney capsule. The ease of this technique reduces stress to the cells and the mouse.
