Cell Purification Method

A cell purification method is a laboratory technique used to isolate a desired cell population from a mixed sample, improving the consistency and interpretability of biological research. These methods exploit differences in cellular properties such as size, density, surface markers, or attachment behavior through processes including centrifugation, selective adhesion, antibody-based separation, and flow sorting. In neuroscience, purified neuronal, glial, or progenitor populations support studies of development, signaling, disease mechanisms, and treatment responses. By reducing unwanted cellular variation, cell purification strengthens downstream analyses and helps researchers link molecular or functional changes to specific cell types.

Quantification of cellular inflammation in the injured/pathological CNS by flow cytometry is complicated by lipid/myelin debris that can have similar size and granulation to cells, decreasing sensitivity/accuracy. We have advanced a cell preparation method to remove myelin debris and improve cell detection by flow cytometry in the injured spinal cord.

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study. In this video, a step-by-step generalized procedure is described for how to perform plasmid purification.

We describe here a novel, robust, and efficient tandem affinity purification (TAP) method for the expression, isolation, and characterization of protein complexes from eukaryotic cells. This protocol could be utilized for the biochemical characterization of discrete complexes as well as the identification of novel interactors and post-translational modifications that regulate their function.
