Cover Slip Transfection

Cover slip transfection is a laboratory method for introducing DNA or RNA into cells cultured on glass coverslips, enabling gene manipulation in samples prepared for microscopy and other analyses. In a typical workflow, nucleic acids associate with a transfection reagent to form complexes that contact the cells, enter them, and release genetic material for transient expression or gene silencing. In neuroscience, the method is especially useful for studying neuronal morphology, protein localization, signaling, and cellular responses at single-cell resolution. Because transfected neurons remain on optically accessible coverslips, researchers can combine genetic manipulation with fluorescence imaging and functional assays.

Rolling Without Slipping

People have observed the rolling motion without slipping ever since the invention of the wheel. For example, one can look at the interaction between a car's tires and the surface of the road. If the driver presses the accelerator to the floor so that the tires spin without the car moving forward, there must be kinetic friction between the wheels and the road's surface. If the driver slowly presses the accelerator, causing the car to move forward, the tires roll without slipping. It is essential...

Rolling with slipping is a physical phenomenon that occurs when a rolling object experiences both rotational and linear motion but also experiences frictional forces that cause slipping. This phenomenon can occur in various situations, such as when a tire rolls on a wet road or a ball rolls on a rough surface. An object's rolling motion is characterized by its rotation around its axis, while linear motion refers to the object's translational motion along a surface. Frictional forces can affect...

An Introduction to Transfection

2023

Transfection is the process of inserting genetic material, such as DNA and double stranded RNA, into mammalian cells. The insertion of DNA into a cell enables the expression, or production, of proteins using the cells own machinery, whereas insertion of RNA into a cell is used to down-regulate the production of a specific protein by stopping translation. While the site of action for transfected RNA is the cytoplasm, DNA must be transported to the nucleus for effective transfection. There, the...

Neuronal Transfection Methods

Transfection - the process of transferring genetic material into cells - is a powerful tool for the rapid and efficient manipulation of gene expression in cells. Because this method can be used to silence the expression of specific proteins or to drive the expression of foreign or modified proteins, transfection is an extremely useful tool in the study of the cellular and molecular processes that govern neuron function. However, mature neurons have a number of properties that make them...
