Dab Immunohistochemistry

DAB immunohistochemistry is a tissue-labeling method that uses 3,3′-diaminobenzidine (DAB) to visualize specific proteins or cellular structures, making it valuable for examining nervous system organization. In the assay, a primary antibody binds the target antigen, a secondary antibody linked to horseradish peroxidase attaches to the primary antibody, and hydrogen peroxide oxidizes DAB to form an insoluble brown precipitate at the binding site. Researchers use this contrast to map neuronal markers, glial proteins, and disease-associated changes in brain and spinal cord sections. The method supports microscopic analysis of cell distribution, morphology, and protein expression in neuroscience research.

Neural circuits are topographically organized into functional compartments with specific molecular profiles. Here, we provide the practical and technical steps for revealing global brain topography using a versatile wholemount immunohistochemical staining approach. We demonstrate the utility of the method using the well-understood cytoarchitecture and circuitry of cerebellum.
