Horizontal Brain Slice

Horizontal brain slices are ex vivo sections of brain tissue cut parallel to the base of the brain, preserving interconnected neural structures within a flat preparation for study. Researchers typically remove and rapidly section brain tissue in chilled, oxygenated artificial cerebrospinal fluid, then maintain the slice under controlled conditions while neurons and synapses remain physiologically responsive; access to the exposed surface enables electrophysiological and imaging measurements. In neuroscience, this preparation supports analysis of circuit connectivity, synaptic transmission, pharmacological responses, and disease-related changes, helping researchers link cellular mechanisms with activity across anatomically defined pathways.

2025

This video demonstrates a detailed protocol for preparing horizontal hippocampal brain slices from mice, focusing on preserving the integrity of hippocampal fiber pathways. The process involves carefully bisecting and securing the brain, using a vibratome for slicing, and maintaining optimal tissue conditions with a carbogenated fluid, resulting in hippocampal brain slices ready for further experimentation.

This article aims to describe a systematic protocol to obtain horizontal hippocampal brain slices in mice. The objective of this methodology is to preserve the integrity of hippocampal fiber pathways, such as the perforant path and the mossy fiber tract to assess dentate gyrus related neurological processes.

We developed a horizontal slice preparation of the adult mammalian retina with the plane of sectioning parallel to the retinal surface. Dendritic fields of nonradially oriented retinal neurons were not truncated, allowing the study of signal processing with patch-clamp and imaging techniques.

Preparing Wedge Brain Slices to Visualize Auditory Neurons

This video demonstrates the process of preparing wedge-shaped brain slices in a laboratory setting. Using a vibratome, a wedge-shaped slice from a mouse's brain is crafted with varying thickness. This slice contains the auditory nerve root positioned on the thicker end, while the hearing-related neurons are located on the thinner end.
