In Vivo Brain Scanning

In vivo brain scanning comprises imaging methods that measure brain structure, activity, blood flow, or molecular signals in living organisms. It matters because it enables researchers and clinicians to study neural systems without removing tissue. Magnetic resonance imaging uses magnetic fields and radiofrequency pulses, while functional MRI detects blood oxygenation changes associated with neural activity; PET uses radiolabeled tracers to map metabolism or receptor distribution. These measurements support research on cognition, development, neurological disease, and treatment response, while longitudinal scans can track changes over time. Combining modalities with behavioral or electrophysiological data improves links between brain mechanisms and observable behavior.

Mitochondrial visualization and analysis from mammalian brain tissue is a challenging task. Here, we describe how three dimensional (3D) reconstruction analysis from the serial block-face scanning electron microscopy (SBFSEM) can be used to gain insights on the morphological and volumetric analysis of this critical energy generating organelle.

Single-cell electroporation (SCE) is a specialized technique allowing delivery of DNA or other macromolecules into individual cells within intact tissue, including in vivo preparations. Here we detail the procedure for SCE of a fluorescent dye or plasmid DNA into neurons within the intact brain of the Xenopus laevis tadpole.
