Interstitial Cell Lineage

Interstitial cell lineage describes the developmental history of interstitial cells, tracing how progenitors divide, differentiate, and generate specialized descendants within nervous system tissues. Researchers reconstruct this lineage by labeling precursor populations and tracking inherited markers, while comparing gene expression, cell morphology, and position across developmental stages. In neuroscience, these approaches clarify how neural and supporting cell populations arise, migrate, and integrate into tissue architecture. Lineage analysis can reveal sources of cellular diversity, identify developmental disruptions linked to neurological disease, and guide studies of neural repair, stem-cell differentiation, and tissue engineering.

Interstitial fluid flow is elevated in solid tumors and can modulate tumor cell invasion. Here we describe a technique to apply interstitial fluid flow to cells embedded in a matrix and then measure its effects on cell invasion. This technique can be easily adapted to study other systems.

This protocol delineates a way to label and trace the fate of small groups of cells zebrafish embryos using UV-uncaging of caged fluorescein, followed by whole mount immunolabeling to amplify the signal from the uncaged fluorescein.

Calcification Assay to Measure Calcium Concentration in Aortic Valve Interstitial Cells

2025

This video demonstrates an assay technique for assessing the calcification of the aortic valve interstitial cells in vitro. The assay helps to quantify the calcium concentration using the arsenazo III reagent, a calcium-sensitive dye that reacts with calcium ions to form a purple-colored calcium-arsenazo complex.

Spatial Measurement of Tumor Interstitial Fluid Pressure: A Method to Measure the Interstitial Fluid Pressure

2023

This video describes a technique of measuring the interstitial fluid pressure within a tumor by perfusion imaging using an image-guided robotic system. Measurements are compared to the intra-tumoral accumulation of liposomes, within a solid tumor.

Here, we present a protocol for a mosaic labeling technique that permits the visualization of neurons derived from a common progenitor cell in two distinct colors. This facilitates neural lineage analysis with the capability of birth-dating individual neurons and studying gene function in the same neurons of different individuals.
