Mounting Media

Mounting media are substances used to preserve and secure biological specimens between a microscope slide and coverslip for imaging and long-term storage. They surround tissue sections, reduce optical distortion by providing a controlled refractive environment, and may protect fluorescent or chromogenic labels from fading or chemical damage. In neuroscience, mounting media help maintain the structure of brain and spinal cord sections while enabling visualization of neurons, glial cells, axons, and synaptic markers through bright-field, fluorescence, or confocal microscopy. Selecting a medium with suitable viscosity, curing properties, refractive index, and label compatibility improves image quality and supports reliable analysis of neural tissue.

Here, we describe a protocol for whole mount immunofluorescence and image-based quantitative volumetric analysis of early stage mouse embryos. We present this technique as a powerful approach to qualitatively and quantitatively assess cardiac structures during development, and propose that it may be widely adaptable to other organ systems.
