Optical Density Calibration

Optical Density Calibration is the process of establishing a reliable relationship between measured light intensity and the concentration or transparency of a sample. It typically uses reference standards or blank measurements to correct instrument response, while the Beer-Lambert principle links absorbance to the amount of light-absorbing material along an optical path. In neuroscience, calibration supports quantitative analysis of stained tissue, neural cultures, and spectrophotometric or imaging-based assays by reducing measurement bias and improving comparability across samples and experiments. Well-calibrated optical density data can strengthen assessments of protein expression, cellular structure, and changes associated with neural development or disease.
